The meaning is obvious: happy birthday. It is a banmal or informal greeting. You may say it to congratulate your friends. Jukyung (Moon Ga Young) is not really close to Suho (Cha Eun Woo) but they are in the same age. So, it is fine if Jukyung speaks informally to Suho.
